Dry powder inhalation is an effective method for delivering a wide range of drug substances to treat chronic obstructive pulmonary diseases. The development of dry powder inhalation products requires a holistic consideration of the device and the formulation. A thorough understanding of the behavior of dry powders during dry powder inhalation product development and usage is crucial in design and developing robust dry powder inhalation products. In this chapter, a systematic discussion of various modeling tools, including the discrete element methods, coupled discrete element methods with computational fluid dynamics, and coupled computational fluid dynamics with the discrete particle method, is presented. How these computational tools can be used to enhance our understanding of dry powder inhalation, such as agglomeration, de‐agglomeration, dispersion, and deposition, is explored. It is anticipated that the advances in computational modeling of dry powder inhalation would enable a sustainable development and manufacturing of dry powder inhalation products.
